Mannitol Salt Agar (MSA) is a widely used selective and differential culture medium designed for the isolation and identification of Staphylococcus aureus in clinical microbiology, food safety testing, pharmaceutical quality control, and laboratory bacterial research. The high sodium chloride concentration supports the growth of salt-tolerant staphylococci while inhibiting most other bacteria, making Mannitol Salt Agar an essential tool for Staphylococcus screening and pathogen detection.

This mannitol fermentation agar contains D-mannitol and phenol red indicator, enabling rapid differentiation of Staphylococcus aureus. When cultured on Mannitol Salt Agar, S. aureus ferments mannitol and produces yellow colonies with a yellow surrounding zone, a typical reaction widely used in microbiology laboratories and bacterial identification workflows.

Each liter of Mannitol Salt Agar medium includes pancreatic digest of casein 5.0 g, peptic digest of animal tissue 5.0 g, beef extract 1.0 g, D-mannitol 10.0 g, sodium chloride 75.0 g, agar 15.0 g, and phenol red 0.025 g, with a final pH of 7.4 ±0.2 at 25°C. To prepare the medium, dissolve 111.0 g of Mannitol Salt Agar powder in 1000 ml purified water, heat with stirring, boil for 1 minute, and sterilize at 121°C for 15 minutes.
