The Urea Broth formulation includes pancreatic digest of casein, potassium dihydrogen phosphate, dextrose, sodium chloride, urea, and phenol red, which together create an optimal environment for bacterial growth. When bacteria metabolize urea, ammonia is produced, resulting in an alkaline pH and a visible color shift to red-pink, indicating urease production and facilitating identification.
With a final pH of 6.9±0.2 at 25°C, this medium is perfectly suited for isolating and identifying key Enterobacteria. It is widely used in clinical microbiology to differentiate Proteus species from other pathogens, such as Salmonella and Shigella.
