Vogel-Johnson Agar Medium for Staphylococcus aureus Detection (USP Standard)

Vogel-Johnson Agar Medium is a highly selective microbiological culture medium designed for the isolation and identification of Staphylococcus aureus in pharmaceutical laboratories, food safety laboratories, and clinical microbiology testing environments. Many microbiology laboratories use Vogel-Johnson Agar Medium for USP microbiological testing, pathogen detection, microbial contamination analysis, and food microbiology quality control, because the medium provides clear differentiation of coagulase-positive staphylococci.

The formulation of Vogel-Johnson Agar Medium contains gelatin peptone and yeast extract, which supply essential nitrogen sources, amino acids, vitamins, and microbial growth factors that promote bacterial growth. Mannitol acts as a fermentable carbohydrate used in bacterial identification testing, while phenol red functions as a pH indicator to visualize acid production. Lithium chloride and glycine suppress competing microorganisms, allowing selective growth of Staphylococcus aureus. When potassium tellurite is added, Vogel-Johnson Agar Medium enables Staphylococcus aureus to reduce tellurite and form black colonies surrounded by yellow zones, which supports reliable microbial identification, pathogen isolation, and laboratory culture analysis.

Preparation Method

Dissolve 61.0 g of Vogel-Johnson Agar Medium powder in 1000 ml purified water, boil for one minute to ensure complete dissolution, and sterilize by autoclaving at 121°C for 15 minutes. Cool the medium to 45–50°C, add 20 ml of sterile filtered 1% potassium tellurite solution, mix thoroughly, and pour into sterile Petri dishes for selective bacterial culture and microbiological testing.

Colony Characteristics

Under 30–35°C aerobic incubation for 24–48 hours, Staphylococcus aureus typically produces black colonies with yellow zones, while other bacteria show inhibited growth or different colony characteristics. This differentiation helps laboratories perform accurate Staphylococcus aureus detection, food safety microbiology testing, and pharmaceutical microbial limit testing.
